Linked Marker Assay Development

Using AFLP ® fingerprints and phenotypic data provided by the customer, genetic markers closely linked to the trait of interest are selected. (A linked marker is a specific band from a fingerprint, which is only present when a specific trait is present in a plant. This band represents a genetic sequence in the neighborhood of the gene coding for this trait). The identified linked markers are tested on a wide range of varieties to determine the predictive value of the assay. The AFLP ® markers are then converted into simple to use PCR assays, which will enable high-throughput in-house testing of plants and seeds.

Its Value
Linked marker assays:

  • enable faster product development;
  • allow selections at seedling-stage;
  • are an alternative to time consuming (infectious) bio-tests which require specific circumstances;
  • give higher accuracy of selection;
  • are suitable for the testing of both monogenetic as well as polygenetic traits;
  • provide detailed knowledge regarding the genetic location of the trait.

EXAMPLE
KeyGene has developed over a hundred simple marker assays that are now being used for economically important traits such as fruit color, and resistance to insects, fungi and nematodes. The attached KeyFact illustrates the linked marker assay development procedure:
