The invented relates to an improved protocol for preparing a sequence library, which involves duplex sequencing of covalently linked DNA strands. This method is of great benefit to long-read sequencing. It enhances the accuracy of sequencing by creating consensus reads per duplex, thereby improving the reliability of sequencing results.
